The coronavirus disease-19 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is characterized by a mild-to-moderate respiratory syndrome, resulting in severe pneumonia and even death. Here, we developed a recombinant receptor-binding domain (RBD)-based subunit vaccine of SARS-CoV-2 linked with a novel cell-penetrating peptide. Sera from mice immunized with RBD-KP2 had higher RBD-specific immunoglobulin G levels as well as superior and more persistent neutralizing activity against SARS-CoV-2 than sera from mice immunized with RBD alone. Most importantly, unlike general recombinant subunit vaccines, RBD-KP2 induced both enhanced CD4+ and CD8+ T cell responses that are crucial for immunogenicity, suggesting that RBD-KP2 is a potentially safe and effective recombinant subunit vaccine against SARS-CoV-2. In addition, RBD-KP2 immunization had completely protective activity against SARS-CoV-2 infection using K18 hACE2 transgenic mice. Our findings also provide a template for designing subunit vaccines for other diseases.
