• 제목 : Serial Femtosecond Crystallography (SFX) : New hope for biological sciences comprising structural biology, protein dynamics, and structure-guided drug discovery/development

Because three-dimensional protein structure can provide important information, structure-based drug discovery and development possibly leads a way for researchers to find a drug lead using rational approaches. In addition, the introduction of the X-ray free-electron laser (XFEL) opened a new realm of the extreme peak brightness X-ray sources with few-femtosecond pulse duration and full transverse coherence. At the Pohang Accelerator Laboratory XFEL (PAL-XFEL), we currently demonstrated an HXRSS FEL using an FBD monochromator that has favorable source features compared to SASE mode: a high average peak spectral intensity exceeded that of SASE by a factor of 12 in self-seeded mode; substantial improvements in the stability of self-seeding; and substantially suppressed pedestal effects. Here, I will show the way how we can prepare for the SFX experiment, collect the data, and calculate them for obtaining the electron density map of a specific protein. By utilizing XFEL, we can get a protein structure at room temperature without radiation damage, leading us to get a better scaffold at the initial stage of the drug discovery program.
